Product Rinse Sterility Test
Combination Product: The product rinse sterility test is specifically designed for products with hollow tubes, such as infusion and infusion sets, which are unsuitable for soaking, and where the fluid channel maintains its sterile state. This method simplifies the process by necessitating the modification of FTM media with liquid D rinse products. Subsequently, the eluate undergoes membrane filtration and is placed in FTM and SCDM. Generally, this method is not the standard choice.


Bulk Drugs / Biopharmaceuticals

Bulk biologics are subjected to sterility testing as outlined in 21 CFR 610.12. This involves utilizing a medium (FTM) with specified sample test sizes, with a minimum capacity of 10 ml, as detailed in the document.


Interpretation of Sterility Test Results
Technicians receive comprehensive training in growth testing methodologies during the cultivation phase. Growth is assessed by examining the medium's transparency against a light source, where any turbid (cloudy) areas signify microbial presence. Upon detecting growth, further testing is conducted on the suspect container to verify that the turbidity is microbial rather than due to sample decomposition. Occasionally, cloudiness may result from particle shedding or chemical reactions with the medium. After testing, samples are returned to the incubator to complete the incubation period. Turbid samples within the medium are repositioned and monitored for an additional four days on day 14 of the test.
